The 25-year-old challenger was stopped by referee Howard Foster in the ninth round, after looking in danger for the first time in the fight.
Groves was well in front on the scorecard, knocking the champion down in the first round, for only the second time in his career, before the stoppage.
Both fighters have insisted they would welcome a rematch, with the 20,000-strong Manchester crowd backing the suggestion.
